Introduction

GoldLink kits have been developed for early stage R&D where users have to analyze the viability of a range of antibodies before optimization of the conjugate on a large scale.

The GoldLink kits enable screening of the preferred antibodies in-house economically by the user, before actually availing BBI’s Custom Conjugation Service to advance their development. In this way, R&D time will be streamlined and efficient use of budget ensured while establishing at the early stage which antibodies are suitable for the product. The kits, comprising BBI’s unique recipe gold colloid are stable to high salt buffers and are previously activated for antibody labeling. The antibodies are conjugated to the functionalized gold via the lysine residues, which offer a non-reversible stable linkage. Antibody pre-treatment is not required. A single reaction buffer is used to conjugate all the antibodies. The process requires minimal antibody usage and does not need a purification step which implies time and cost savings.

Proteins or antibodies are attached covalently to highly stable BBI GoldLink colloids at an extremely high OD rapidly and easily. The kit procedure’s hands-on-time is only 2 minutes and the conjugate can be used in 15 minutes. A protective surface coat is provided on the gold colloid, which can withstand the most extreme conditions such as 1M NaCl or 2.5M NaOH at 70° C for >1 hour. The GoldLink colloid in the kits is supplied as a freeze dried mixture. As elaborated in Fig.1, the level of conjugate sensitivity achieved through the use of a GoldLink kit is suitable for antibody screening during initial R&D.

Procedure Using the GoldLink Rapid Conjugation Kit

The 15-minute procedure involves the following steps:

  • The antibody is diluted
  • It is then mixed with a reaction buffer
  • It is then added to freeze dried BBI gold colloid
  • The reaction is then quenched
  • The finished conjugate is then obtained

For Custom Conjugation of Commercial Scale Up

Customers who want to achieve a higher level of sensitivity and/or a fully optimized conjugate, BBI also offers a Custom Conjugate Service. BBI will take the antibody of choice and using its long established, proven processes, optimize the conjugate, and then freeze the specific recipe and put a commercial supply plan in place.

BBI is capable of assisting all stages of conjugation from initial R&D antibody screening to commercial product supply.

Firstly, Goldlink kits are used in-house with initial R&D pre-optimization screening. Next in the custom conjugation service, BBI will optimize the selected antibodies offering further sample conjugate. The conjugate will then be optimized by BBI for sensitivity, specificity, validation and stability. Finally robust, validated conjugate in a single batch from 100 ml up to 35L OD10 will be supplied by BBI.
