Contractility Under Physiological Conditions—FLEXcyte 96

The FLEXcyte 96 available from Nanion Technologies takes in vitro contraction force measurements of cardiac cells to a new level. The system employs a well-defined, flexible substrate for cardiac cells, thus leveraging mature responses and maturity of stem cell-derived cardiomyocytes to pharmacological compounds in a larger throughput format.

The cells present in the 96-well plate bind to the flexible substrates as monolayers and the contractility of the synchronized cell layers are recorded. This special technology enables recordings to be performed in a native-like surrounding simulating the mechanical conditions of the heart.

The FLEXcyte 96 is a ready-to-run system for functional and structural contraction force recordings in efficacy and safety screening, fundamental research, and cardiotoxicity.

The FLEXcyte 96 is available as a supplement to the CardioExcyte 96. The polydimethylsiloxane (PDMS) membranes of the FLEXcyte 96 disposable plates have a thickness of <10 μm and include state-of-the-art surface modification, thus providing the strong mechanical support and physiological elasticity of native human heart tissue.

Upon being deflected due to the weight of the culture medium, rhythmic contraction of the cardiomyocytes raises the membranes in the 96-well in the upward direction. The mechanical stress calculation can be done by measuring the variations in deflection.

The system has been verified with stem cell-derived cardiomyocytes from a number of providers (Cellular Dynamics International, NEXEL, Axol, and Ncardia). Human-induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) exhibit positive inotropic responses, thereby avoiding the need for further stimulation of “cell maturation.” The force of cardiac contraction can also be quantified on patient-derived cells.

Without missing any subtleties of cytotoxic responses, contractile forces on heart muscle cells can be reliably measured for efficacy, toxicity, and safety analyses in both acute and chronic time frames. The FLEXcyte 96 is a completely automatic device, recording from 96 wells at the same time.
