This video from JPK Instruments shows an optical trap which can be used to pull membrane tethers from live cells. The CHO cells are frozen on to a glass substrate and brought in contact with an optically trapped microsphere, which is coated with ligands specifically bind to the cell surface. The speed of pulling depends on the contact time.
